Product Overview
This rabbit polyclonal antibody recognizes TRAF2 and NCK-interacting protein kinase (gene TNIK) in human and mouse samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 154.9 kDa; the supplier reports observed bands at 180,150 kDa.
TNIK is a serine/threonine kinase of the germinal center kinase family that is required for canonical Wnt target gene activation, acting at promoters bound by TCF4/TCF7L2 and β-catenin. It also influences JNK signaling, cytoskeletal organization, neuronal dendrite growth and Hippo pathway activation, and it is studied in Wnt-driven cancers such as colorectal cancer.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human, Mouse) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human K562 cells, human HEL cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human cervix squamous cells carcinoma tissue, human placenta tissue, human prostate adenocarcinoma tissue, mouse brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): PC-3 cells.
Samples with a confirmed signal (flow cytometry): K562 cells.
Immunogen
Recombinant human TNIK fragment (His553–Glu886), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places TRAF2 and NCK-interacting protein kinase in the nucleus, cytoplasm and recycling endosome. Tissue expression noted by UniProt (human): Expressed ubiquitously. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
