Product Overview

This rabbit polyclonal antibody detects TNFAIP3-interacting protein 1 (gene TNIP1) in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 71.9 kDa, and the supplier reports an observed band at about 80–85 kDa.

ABIN-1 (TNIP1) is an A20-binding protein that limits NF-κB activation by helping the deubiquitinase A20 (TNFAIP3) act on the ubiquitinated regulator NEMO (IKBKG). It also dampens ERK signaling, shapes macrophage inflammatory responses and helps prevent autoimmunity, and genetic variants near TNIP1 have been associated with psoriasis and lupus.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human HEK293 cells, human Raji cells, human A549 cells, human Caco-2 cells, human T-47D cells, rat spleen tissue, rat lung tissue, mouse lung tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant human TNIP1 fragment (Glu312–Lys371), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places TNFAIP3-interacting protein 1 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 71.9 kDa; observed bands at about 80 and 85 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human TNIP1 fragment (Glu312–Lys371), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
