Product Overview
This rabbit polyclonal antibody recognizes troponin I, slow skeletal muscle (gene TNNI1) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 21.7 kDa, and the supplier reports an observed band at about 22 kDa.
Troponin I slow skeletal (TNNI1) is the inhibitory subunit of troponin in slow-twitch skeletal muscle. It blocks actomyosin ATPase activity at rest and releases this block when calcium binds troponin C, making contraction calcium-dependent. TNNI1 is also expressed in the fetal heart and serves as a marker of slow muscle fibers and immature cardiomyocytes.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
Samples with a confirmed band (WB): human HepG2 cells, human U-87MG cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human skeletal muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the N-terminal region of human TNNI1. Sequence identity with the mouse and rat orthologs: 94.7%.
Reactivity Notes
The supplier lists reactivity with human. Tissue expression noted by UniProt (human): Highest levels observed in human skeletal muscle (e.g. gastrocnemious muscle), differentiated cultures of primary human muscle cells and rhabdomyosarcoma cells cultured in low serum medium. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
