Product Overview
This rabbit polyclonal antibody recognizes toll-like receptor 4 (gene TLR4) in human samples and is validated for Western blot and IHC. The predicted molecular weight is 95.7 kDa, and the supplier reports an observed band at about 80 kDa.
Toll-like receptor 4 (TLR4) is the receptor for bacterial lipopolysaccharide (LPS), working with the co-receptor MD-2 (LY96) and CD14. It signals from the cell surface through MyD88 and, after internalization, through TRIF, activating NF-κB, IRF3 and inflammatory cytokine production. TLR4 is central to research on sepsis, endotoxin responses and sterile inflammation.

Validated Applications
| IHC (paraffin sections) | 0.5–1 µg/mL (Human) |
|---|---|
| Western blot | 0.1–0.5 µg/mL (Human) |
Samples with a confirmed band (WB): HeLa cells, SMMC cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Samples with confirmed staining (IHC): human lung cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide from the N-terminal region of human TLR4.
Reactivity Notes
The supplier lists reactivity with human. UniProt places toll-like receptor 4 in the cell membrane, early endosome and cell projection. Tissue expression noted by UniProt (human): Highly expressed in placenta, spleen and peripheral blood leukocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
