Overview
Antibody against TP53 (Cellular tumor antigen p53) for research detection of protein expression and localization.
Target biology
TP53 encodes p53, a stress-responsive transcription factor that integrates DNA damage, oncogenic stress and other insults to induce cell-cycle arrest, senescence or apoptosis. p53 regulates genes involved in DNA repair, metabolism and cell fate decisions and is among the most frequently altered tumor suppressors in cancer.
Expected localization
Predominantly nuclear, with regulated cytoplasmic localization depending on stress and post-translational modifications.
Research context
This target is frequently studied in the context of Oncology & Angiogenesis research. Application suitability (e.g., ELISA, IHC, WB) is indicated in the specifications; conditions should be optimized by titration. Species reactivity information is provided in the specifications (reported: Human).
Recommended experimental notes
- For Western blotting, optimize lysis conditions and include reducing/non-reducing conditions as appropriate; titrate antibody to balance sensitivity and background.
- For IHC, fixation and antigen retrieval strongly influence epitope accessibility; test citrate- vs EDTA-based retrieval and include matched positive/negative tissues.
- For ELISA, ensure capture/detection antibody compatibility and run a standard curve; verify sample matrix effects with spike-and-recovery and dilution linearity.
Controls & interpretation
Where possible, confirm specificity using genetic perturbation (KO/KD/CRISPR), peptide competition (if applicable), or an orthogonal readout. Include a negative control sample expected to express low/none of TP53, and a positive control sample known to express TP53.
Immunogen context
Generated using an immunogen derived from E. coli - derived recombinant Human TP53/p53 (Asp7-Asp393).. When interpreting results, note that epitope accessibility can vary with denaturation, fixation, or post‑translational modifications; empirical optimization is recommended.
