Product Overview
This rabbit polyclonal antibody detects thiopurine S-methyltransferase (gene TPMT) in human, mouse and rat samples and is validated for ELISA and Western blot. The predicted molecular weight is 28.2 kDa, and the supplier reports an observed band at about 28 kDa.
Thiopurine S-methyltransferase (TPMT) methylates thiopurine drugs such as 6-mercaptopurine and 6-thioguanine using S-adenosylmethionine, which lowers the amount converted into active thioguanine nucleotides. Inherited low TPMT activity increases the risk of severe toxicity from these drugs, making TPMT a classic example in pharmacogenetics.

Samples with a confirmed band (WB): human SGC-7901 cells, rat testis tissue, mouse testis tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places thiopurine S-methyltransferase in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
