Rabbit Monoclonal

Product Overview

Clone 23T27 is a rabbit monoclonal antibody that recognizes tax1-binding protein 1 (gene TAX1BP1) in human, mouse and rat samples and is validated for Western blot and ICC/IF. The predicted molecular weight is 90.9 kDa, and the supplier reports an observed band at about 91 kDa.

TAX1BP1 is a ubiquitin-binding adaptor that works with the deubiquitinase A20 (TNFAIP3) to switch off NF-κB and IRF3 signaling after inflammatory or antiviral stimulation. It also acts as a selective autophagy receptor that helps clear protein aggregates and intracellular bacteria, and it was first identified as a binding partner of the HTLV-1 Tax protein.

Validated Applications

Western blot 1:500–1:2000
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human HepG2 cells, human RT4 cells, human SIHA cells, rat testis tissue, mouse testis tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human TRAF6BP.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places tax1-binding protein 1 in the cytoplasm, mitochondrion, preautophagosomal structure and cytoplasmic vesicle. Tissue expression noted by UniProt (human): Expressed in all tissues tested. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
