Product Overview
This rabbit polyclonal antibody recognizes CD40 ligand (gene CD40LG) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 29.3 kDa, and the supplier reports an observed band at about 36 kDa.
CD40 ligand (CD40LG, also called CD154) is a TNF family protein expressed mainly on activated CD4+ T cells and platelets. It binds CD40 to drive B-cell activation, immunoglobulin class switching and dendritic cell maturation, and loss of function causes X-linked hyper-IgM syndrome.

Samples with a confirmed band (WB): human HL-60 cells, human K562 cells, human Raji cells, human Jurkat cells, rat RH35 cells, mouse spleen tissue, mouse ANA-1 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): C6 cells, human PBMC cells.

Recombinant human TRAP/CD40L/CD40LG fragment (Glu76–His249), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places CD40 ligand in the cell membrane and cell surface. Tissue expression noted by UniProt (human): Specifically expressed on activated CD4+ T-lymphocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
