Overview

This antibody is intended for detection of TERF2 (Telomeric repeat-binding factor 2) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-TRF2/TERF2 Antibody Picoband® catalog # A00650-1. Tested in IHC, WB applications. This antibody reacts with Human, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E.coli-derived human TRF2 recombinant protein (Position: A81-K287). Human TRF2 shares 95.7% amino acid (aa) sequence identity with mouse TRF2. (reported region: A81-K287).
  • Molecular weight context: reported MW: 75 kDa; calculated MW: 59594 MW
  • Reactivity: Human,Rat
  • Applications: IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Telomeric repeat-binding factor 2; Telomeric repeat-binding factor 2. Telomeric repeat-binding factor 2, also known as TERF2, TRF2, or TRBF2, is encoded in humans by the TERF2 gene. This gene encodes a telomere specific protein, TERF2, which is a component of the telomere nucleoprotein complex. This protein is present at telomeres in metaphase of the cell cycle, is a second negative regulator of telomere length and plays a key role in the protective activity of telomeres. While having similar telomere binding activity and domain organization, TERF2 differs from TERF1 in that its N terminus is basic rather than acidic. Functional note: Binds the telomeric double-stranded 5'-TTAGGG-3' repeat and plays a central role in telomere maintenance and protection against end-to-end fusion of chromosomes. In addition to its telomeric DNA-binding role, required to recruit a number of factors and enzymes required for telomere protection, including the shelterin complex, TERF2IP/RAP1 and DCLRE1B/Apollo. Component of the shelterin complex (telosome) that is involved in the regulation of telomere length and protection. Shelterin associates with arrays of double-stranded 5'-TTAGGG-3' repeats added by telomerase and protects chromosome ends; without its protective activity, telomeres are no longer hidden from the DNA damage surveillance and chromosome ends are inappropriately processed by DNA repair pathways. Together with DCLRE1B/Apollo, plays a key role in telomeric loop (T loop) formation by generating 3' single- stranded overhang at the leading end telomeres: T loops have been proposed to protect chromosome ends from degradation and repair. Required both to recruit DCLRE1B/Apollo to telomeres and activate the exonuclease activity of DCLRE1B/Apollo. Preferentially binds to positive supercoiled DNA. Together with DCLRE1B/Apollo, required to control the amount of DNA topoisomerase (TOP1, TOP2A and TOP2B) needed for telomere replication during fork passage and prevent aberrant telomere topology. Recruits TERF2IP/RAP1 to telomeres, thereby participating in to repressing homology- ed repair (HDR), which can affect telomere length. . Reported localization: Nucleus . Chromosome, telomere . Colocalizes with telomeric DNA in interphase cells and is located at chromosome ends during metaphase. Expression/tissue context: Ubiquitous. Highly expressed in spleen, thymus, prostate, uterus, testis, small intestine, colon and peripheral blood leukocytes.

Research relevance and current trends

  • Cancer: Researchers commonly examine how TERF2 (Telomeric repeat-binding factor 2) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative TERF2 (Telomeric repeat-binding factor 2) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of TERF2 (Telomeric repeat-binding factor 2) across tissue regions and cell types using matched controls.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.

Is this A00650-1 anti-TRF2/TERF2 antibody reactive to the isotypes of TERF2?
The immunogen of A00650-1 anti-TRF2/TERF2 antibody is E.coli-derived human TRF2 recombinant protein (Position: A81-K287). Human TRF2 shares 95.7% amino acid (aa) sequence identity with mouse TRF2. Could you tell me which isotype you are interested in so I can help see if the immunogen is part of this isotype?
Does A00650-1 anti-TRF2/TERF2 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
It shows on the product datasheet, A00650-1 anti-TRF2/TERF2 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
