Product Overview

This rabbit polyclonal antibody targets triadin (gene TRDN) in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 81.6 kDa, and the supplier reports an observed band at about 25 kDa.

Triadin (TRDN) is a transmembrane protein of the sarcoplasmic reticulum at the triad and dyad junctions of skeletal and heart muscle. It links the ryanodine receptors RYR1 and RYR2 with calsequestrin and helps organize the junction, fine-tuning calcium release during excitation–contraction coupling. TRDN mutations cause inherited cardiac arrhythmia syndromes.

Form Lyophilized
Formulation Per vial: 4 mg trehalose; 0.9 mg NaCl; 0.2 mg Na2HPO4
Calculated MW 81.6 kDa
Observed MW 25 kDa
Storage As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human MCF-7 cells, human K562 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with a confirmed signal (flow cytometry): MCF-7 cells.

Immunogen

Recombinant human Triadin/TRDN fragment (Met67–Gln729), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human. UniProt places triadin in the cell membrane and sarcoplasmic reticulum membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 81.6 kDa; observed band at about 25 kDa. The observed band is much smaller than the calculated size, which can indicate a processed form or a shorter isoform; check the supplier images.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Triadin/TRDN fragment (Met67–Gln729), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
