| Immunogen | A synthesized peptide derived from human TRIB3 Disrupts insulin signaling by binding directly to Akt kinases and blocking their activation. May bind directly to and mask the 'Thr-308' phosphorylation site in AKT1. Binds to ATF4 and inhibits its transcriptional activation activity. Interacts with the NF-kappa-B transactivator p65 RELA and inhibits its phosphorylation and thus its transcriptional activation activity. |

Product Overview
Clone AEIE-20 is a rabbit monoclonal antibody that recognizes tribbles homolog 3 (gene TRIB3) in human samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 39.6 kDa, and the supplier reports an observed band at about 40 kDa.
Tribbles homolog 3 (TRIB3) is a stress-induced pseudokinase that is strongly upregulated during ER stress and the integrated stress response. It inhibits the transcription factor CHOP/ATF4 in a negative feedback loop, can bind and inhibit AKT, and influences insulin signaling and cell death, so it is studied in metabolic disease and cellular stress.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunoprecipitation | 1:20 |
Samples with a confirmed band (WB): human K562 cells, human Caco-2 cells, human 293T cells, human HepG2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human TRIB3.
Reactivity Notes
The supplier lists reactivity with human. UniProt places tribbles homolog 3 in the nucleus. Tissue expression noted by UniProt (human): Highest expression in liver, pancreas, peripheral blood leukocytes and bone marrow. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
