Product Overview
This rabbit polyclonal antibody recognizes E3 ubiquitin-protein ligase TRIM21 in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 54.2 kDa, and the supplier reports an observed band at about 52 kDa.
TRIM21 (Ro52) is a tripartite motif E3 ubiquitin ligase that acts as a high-affinity cytosolic antibody receptor. It binds antibodies attached to viruses that have entered the cell and targets them for proteasomal destruction while triggering innate immune signaling. TRIM21 is also a major autoantigen in Sjögren syndrome and lupus and underlies the Trim-Away protein-depletion technique.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 2 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat lung tissue, mouse lung tissue, rat thymus tissue, mouse thymus tissue, human HeLa cells, human THP-1 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): SiHa cells.
Immunogen
Recombinant human TRIM21/SS-A fragment (Glu12–Tyr475), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places E3 ubiquitin-protein ligase TRIM21 in the cytoplasm, cytoplasmic vesicle and nucleus. Tissue expression noted by UniProt (human): Isoform 1 and isoform 2 are expressed in fetal and adult heart and fetal lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
