Product Overview
This rabbit polyclonal antibody detects E3 ubiquitin/ISG15 ligase TRIM25 in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 71.0 kDa, and the supplier reports an observed band at about 71 kDa.
TRIM25 is a tripartite motif E3 ligase that can attach both ubiquitin and the interferon-induced modifier ISG15. It adds Lys-63-linked ubiquitin chains to the viral RNA sensor RIG-I, promoting type I interferon production, and it binds RNA. TRIM25 is a key protein in antiviral innate immunity research in human and mouse systems.

Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human K562 cells, human Caco-2 cells, human T-47D cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human mammary cancer tissue, human pancreatic cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): U937 cells.
Immunogen
Recombinant human TRIM25/EFP fragment (Met1–Ala610), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places E3 ubiquitin/ISG15 ligase TRIM25 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Expressed in breast tumors (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
