Product Overview
Clone ADFG-20 is a rabbit monoclonal antibody that recognizes E3 ubiquitin/ISG15 ligase TRIM25 in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 71.0 kDa, and the supplier reports an observed band at about 71 kDa.
TRIM25 is a tripartite motif E3 ligase that can attach both ubiquitin and the interferon-induced modifier ISG15. It adds Lys-63-linked ubiquitin chains to the viral RNA sensor RIG-I, promoting type I interferon production, and it binds RNA. TRIM25 is a key protein in antiviral innate immunity research in human and mouse systems.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, mouse stomach tissue, rat stomach tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

The supplier lists reactivity with human, mouse and rat. UniProt places E3 ubiquitin/ISG15 ligase TRIM25 in the cytoplasm and nucleus.
