Product Overview

This rabbit polyclonal antibody recognizes high affinity nerve growth factor receptor (gene NTRK1) in human samples and is validated for Western blot. The predicted molecular weight is 87.5 kDa, and the supplier reports an observed band at about 41 kDa.

TrkA (NTRK1) is the high-affinity receptor tyrosine kinase for nerve growth factor (NGF). NGF binding triggers receptor autophosphorylation and activation of MAPK, PI3K/AKT and PLCγ signaling that supports the survival and differentiation of sensory and sympathetic neurons, and TrkA is studied in pain, neurodevelopment and NTRK fusion-driven cancers.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)

Samples with a confirmed band (WB): COLO320 cells, HT1080 cells, U87 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide from the N-terminal region of human trkA.

Reactivity Notes

The supplier lists reactivity with human. UniProt places high affinity nerve growth factor receptor in the cell membrane, early endosome membrane, late endosome membrane and recycling endosome membrane. Tissue expression noted by UniProt (human): Isoform TrkA-I is found in most non-neuronal tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on zebrafish samples?
A.Reactivity with zebrafish samples has not been tested by the supplier. Aligning the immunogen sequence with the zebrafish ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 87.5 kDa; observed band at about 41 kDa. The observed band is much smaller than the calculated size, which can indicate a processed form or a shorter isoform; check the supplier images.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human trkA.
