Rabbit Monoclonal

Product Overview

Clone 22N08 is a rabbit monoclonal antibody that recognizes BDNF/NT-3 growth factors receptor (gene Ntrk2) in mouse and rat samples and is validated for Western blot, IHC and immunoprecipitation. The predicted molecular weight is 92.1 kDa, and the supplier reports an observed band at about 120–140 kDa.

TrkB (NTRK2) is a receptor tyrosine kinase activated mainly by BDNF and neurotrophin-4. Its signaling supports neuron survival, differentiation and synapse formation and is central to activity-dependent synaptic plasticity, so TrkB is widely studied in learning and memory, depression, neurodegeneration and obesity.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunoprecipitation 1:50

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human trkB.

Reactivity Notes

The supplier lists reactivity with mouse and rat. UniProt places BDNF/NT-3 growth factors receptor in the cell membrane, endosome membrane, early endosome membrane and cell projection. Tissue expression noted by UniProt (mouse): Expressed in the brain, in neurons (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 92.1 kDa; observed bands at about 120 and 140 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human trkB.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
