Product Overview

This rabbit polyclonal antibody detects NT-3 growth factor receptor (gene NTRK3) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 94.4 kDa, and the supplier reports an observed band at about 85–91 kDa.

TrkC (NTRK3) is the receptor tyrosine kinase for neurotrophin-3. Ligand binding triggers autophosphorylation and activation of PI3K/AKT and MAPK signaling that supports cell survival and differentiation, and TrkC is studied in nervous system development, proprioceptive neurons and NTRK fusion-positive tumors.

Validated Applications

IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse)
Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue, U87 cells, SHG cells, NEURO cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human mammary cancer tissue, rat brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide from the N-terminal region of human trkC. It differs from the mouse and rat sequences at one residue.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places NT-3 growth factor receptor in the membrane. Tissue expression noted by UniProt (human): Widely expressed but mainly in nervous tissue. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 0.5–1 µg/mL (Human, Rat, Mouse). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 94.4 kDa; observed bands at about 85 and 91 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the N-terminal region of human trkC. It differs from the mouse and rat sequences at one residue.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
