Product Overview

Clone 21T11 is a rabbit monoclonal antibody that recognizes short transient receptor potential channel 1 (gene TRPC1) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 91.2 kDa, and the supplier reports an observed band at about 120 kDa.

TRPC1 is a member of the canonical transient receptor potential (TRPC) family of cation channels that let calcium and sodium enter cells after receptor stimulation. It often assembles with TRPC4 or TRPC5 into heteromeric channels with altered properties, and it is studied in store-operated and receptor-operated calcium entry in many cell types.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): human PC-3 cells, human HepG2 cells, human Caco-2 cells, human U251 cells, rat testis tissue, mouse lung tissue, mouse testis tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human TRPC1.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places short transient receptor potential channel 1 in the cell membrane. Tissue expression noted by UniProt (human): Seems to be ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 91.2 kDa; observed band at about 120 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human TRPC1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
