Anti-TRPV1 (VR1) (extracellular) Antibody (#ACC-029) is a highly specific antibody directed against an extracellular epitope of the rat TRPV1 channel. The antibody can be used in western blot, immunohistochemistry, and live cell imaging applications. It has been designed to recognize TRPV1 from rat and mouse samples. The antibody will not work with human samples. Anti-TRPV1 (VR1) (extracellular)-ATTO Fluor-488 Antibody (#ACC-029-AG) is directly labeled with an ATTO-488 fluorescent dye. ATTO dyes are characterized by strong absorption (high extinction coefficient), high fluorescence quantum yield, and high photo-stability. The ATTO-488 label is analogous to the well known dye fluorescein isothiocyanate (FITC) and can be used with filters typically used to detect FITC. Anti-TRPV1 (VR1) (extracellular)-ATTO Fluor-488 Antibody has been tested in immunohistochemistry and live cell imaging applications and is especially suited for experiments requiring simultaneous labeling of different markers.

Target TRPV1

Overview

Anti-TRPV1 (VR1) (extracellular)-ATTO Fluor-488 Antibody is an antibody targeting Transient receptor potential cation channel subfamily V member 1, Capsaicin receptor, Vanilloid receptor 1, OTRPC1 Polyclonal raised in Rabbit (ATTO-488. Maximum absorption 501 nm; maximum fluorescence 523 nm. The fluorescence is excited most efficiently in the 480 - 515 nm range. This label is analogous to the dye fluorescein isothiocyanate (FITC) and can be used with filters used to detect FITC.). This antibody is commonly used in FC, IC, IF, IHC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Transient receptor potential cation channel subfamily V member 1, Capsaicin receptor, Vanilloid receptor 1, OTRPC1 (also reported as Transient receptor potential cation channel subfamily V member 1, Capsaicin receptor, Vanilloid receptor 1, OTRPC1).
  • Immunogen/epitope region: 3rd extracellular loop.
  • Homology note: Rat - 14 out of 15 amino acid residues identical Mouse - 12 out of 15 amino acid residues identicalWon't recognize TRPV1 from human samples (informative for cross-species interpretation).
  • Species reactivity (as provided): Rat, Mouse.
  • Specificity statement (as provided): Recognizes TRPV1 from rat only..
  • Cited use: IHC (literature use does not guarantee performance in every setup).
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #ACC-029), and immunohistochemistry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

TRP channels are a large family (about 28 genes) of plasma membrane, non-selective cationic channels that are either specifically or ubiquitously expressed in excitable and non-excitable cells.1According to IUPHAR the TRP family comprises three main subfamilies on the basis of sequence homology; TRPC, TRPM and TRPV (to date, three extra subfamilies are considered to belong to the TRP family; the TRPA, TRPML, and TRPP).1-4 The TRPV subfamily consists of six members, TRPV1-6.5TRPV1 channel has many activators; among them heat, protons, vanilloids like capsaicin, resiniferatoxin (RTX), and lipids. This channel is associated with tissue injury and inflammation.6,7 TRPV1 is expressed predominantly in nociceptors and in sensory neurons.Recent studies demonstrated involvement of TRPV1 in apoptosis where inhibition of the receptor prevented apoptosis.8,9

Research relevance and current trends

  • Linking transporter/channel abundance to ionic homeostasis and excitability-related phenotypes.
  • Studying compartment-specific localization (surface vs intracellular pools) and trafficking dynamics.
  • Combining antibody readouts with functional assays for more complete interpretation.

Common research applications

  • Immunohistochemistry (IHC): examine spatial distribution in tissue and relate signal to cell-type composition.
  • Immunofluorescence/ICC: assess subcellular localization and co-localization with markers in cells or sections.
  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-AG.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.
