Product Overview
This rabbit polyclonal antibody recognizes tRNA-splicing endonuclease subunit Sen34 (gene TSEN34) in human, mouse and rat samples and is validated for Western blot, ICC/IF and ELISA. The predicted molecular weight is 33.7 kDa, and the supplier reports an observed band at about 50 kDa.
TSEN34 is one of two catalytic subunits of the tRNA splicing endonuclease, the complex that removes introns from precursor tRNAs. It is thought to cut at the 3′ splice site, releasing the intron and two tRNA halves that are later joined, and the complex has also been linked to mRNA 3′-end processing. Variants in TSEN genes cause pontocerebellar hypoplasia.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): rat brain tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U87 cells.
Immunogen
Recombinant human TSEN34 fragment (Met1–Gln310), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places tRNA-splicing endonuclease subunit Sen34 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
