Rabbit Monoclonal

Product Overview

Clone 27N58 is a rabbit monoclonal antibody that recognizes homeobox protein Nkx-2.1 (gene NKX2-1) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 38.6 kDa, and the supplier reports an observed band at about 38–42 kDa.

NKX2-1 (also called TTF-1) is an NK2 homeobox transcription factor that drives thyroid-specific genes such as thyroglobulin, thyroid peroxidase and the TSH receptor. It is also needed for lung development and surfactant protein expression, and it is widely used as a lineage marker for thyroid and lung epithelial cells and their tumors.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Samples with a confirmed band (WB): rat lung cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human TTF1.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places homeobox protein Nkx-2.1 in the nucleus. Tissue expression noted by UniProt (human): Thyroid and lung. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 38.6 kDa; observed bands at about 38 and 42 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human TTF1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
