Vendor description (summary): Boster Bio Anti-Tyrosinase/TYR Antibody Picoband® catalog # A00326-3.

Biological background

Biological context: On ligand binding, forms a receptor complex consisting of two type II and two type I transmembrane serine/threonine kinases. Type II receptors phosphorylate and activate type I receptors which autophosphorylate, then bind and activate SMAD transcriptional regulators. Binds to BMP7, BMP2 and, less efficiently, BMP4. Binding is weak but enhanced by the presence of type I receptors for BMPs. Mediates induction of adipogenesis by GDF6.

Expression and localization notes: cellular localization: Cell membrane., tissue context: Highly expressed in heart and liver..

Common research applications

  • Western blotting (WB): Compare TYR levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of TYR in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify TYR-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Background: The enzyme encoded by this gene catalyzes the first 2 steps, and at least 1 subsequent step, in the conversion of tyrosine to melanin. The enzyme has both tyrosine hydroxylase and dopa oxidase catalytic activities, and requires copper for function. Mutations in this gene result in oculocutaneous albinism, and nonpathologic polymorphisms result in skin pigmentation variation. The human genome contains a pseudogene similar to the 3' half of this gene.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cell membrane.
  • Tissue details: Highly expressed in heart and liver.
  • Research category: Cardiovascular,Cytoskeleton/ECM,Extracellular Matrix,Growth Factors/Hormones,Hypertrophy,Mesenchymal Stem Cells,Protein Phosphorylation,Ser/Thr Kinases,Signal Transduction,Signaling Pathways,Stem Cells,Structures,Surface Molecules,TGF Beta
