Product Overview
Clone IAC-21 is a rabbit monoclonal antibody that recognizes ubiquitin-like modifier-activating enzyme 1 (gene UBA1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 117.8 kDa, and the supplier reports an observed band at about 118 kDa.
UBA1 is the major ubiquitin-activating (E1) enzyme that starts the ubiquitination cascade. It uses ATP to activate ubiquitin and forms a thioester with it, then passes ubiquitin to E2 conjugating enzymes, so it is required for protein degradation by the proteasome and for many signaling and DNA repair processes.

Samples with a confirmed band (WB): human Jurkat cells, human SiHa cells, human 293T cells, human PC-3 cells, rat brain tissue, mouse brain tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human acinic cells carcinoma of parotid tissue, human clear cells renal cells carcinoma tissue, human colon adenocarcinoma tissue, human liver cancer tissue, human ovarian cancer tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human UBA1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ubiquitin-like modifier-activating enzyme 1 in the cytoplasm, mitochondrion, nucleus and SUBCELLULAR LOCATION:. Tissue expression noted by UniProt (human): Detected in erythrocytes (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
