Rabbit Monoclonal

Product Overview

Clone 26U65 is a rabbit monoclonal antibody that recognizes ubiquitin carboxyl-terminal hydrolase isozyme L3 (gene UCHL3) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 26.2 kDa, and the supplier reports an observed band at about 26 kDa.

UCH-L3 is a deubiquitinating enzyme of the ubiquitin C-terminal hydrolase family. It processes ubiquitin precursors and removes ubiquitin or the related modifier NEDD8 from small adducts, helping keep cellular ubiquitin levels balanced. It is studied in protein turnover, metabolism and DNA repair.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human K562 cells, human 293T cells, human A431 cells, human Caco-2 cells, rat heart tissue, mouse heart tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.

Samples with confirmed staining (ICC/IF): U2OS cells.

Immunogen

Synthetic peptide based on the sequence of UCHL3.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places ubiquitin carboxyl-terminal hydrolase isozyme L3 in the cytoplasm. Tissue expression noted by UniProt (human): Highly expressed in heart, skeletal muscle, and testis. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
