Product Overview

This rabbit polyclonal antibody detects urokinase plasminogen activator surface receptor (gene PLAUR) in human samples and is validated for Western blot. The predicted molecular weight is 37.0 kDa; the supplier reports observed bands at 39 kDa, 55 kDa.

The urokinase receptor (uPAR, CD87) is a GPI-anchored cell surface receptor that binds uPA and focuses plasminogen activation at the cell surface. It also binds vitronectin and signals through integrins, so it coordinates proteolysis, adhesion and migration, and it is studied in inflammation, cancer invasion and kidney disease.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)

Samples with a confirmed band (WB): HeLa cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Immunogen

Recombinant human PLAUR fragment (Glu230–Ser286), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 63.2%.

Reactivity Notes

The supplier lists reactivity with human. Tissue expression noted by UniProt (human): Expressed in neurons of the rolandic area of the brain (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 37.0 kDa; observed bands at about 39 and 55 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human PLAUR fragment (Glu230–Ser286), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 63.2%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
