Product Overview
This rabbit polyclonal antibody targets regulator of nonsense transcripts 3A (gene UPF3A) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 54.7 kDa, and the supplier reports an observed band at about 51 kDa.
UPF3A is a paralog of UPF3B in the nonsense-mediated mRNA decay (NMD) pathway. It can associate with the exon junction complex and UPF2, but it is generally a weak NMD activator and can even dampen NMD by competing with UPF3B. It is studied in RNA surveillance and in germ-cell and neural development.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human HEK293 cells, rat PC-12 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with a confirmed signal (flow cytometry): HL-60 cells.

Recombinant human UPF3A fragment (Glu4–Glu452), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places regulator of nonsense transcripts 3A in the nucleus and cytoplasm. Tissue expression noted by UniProt (human): Isoform 1 is strongly expressed in testis, uterus, muscle, fetal brain and spinal cord. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
