Product Overview

Clone ADDO-21 is a rabbit monoclonal antibody that recognizes ubiquitin carboxyl-terminal hydrolase 28 (gene USP28) in human samples and is validated for Western blot, ICC/IF and flow cytometry. The predicted molecular weight is 122.5 kDa, and the supplier reports an observed band at about 135 kDa.

USP28 is a deubiquitinating enzyme that protects several proteins from degradation. It stabilizes the MYC oncoprotein by counteracting the ubiquitin ligase FBW7, and it supports DNA damage checkpoint signaling through proteins such as claspin and 53BP1. USP28 is studied in the DNA damage response and cancer.

Validated Applications

Western blot 1:500–1:2000
Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:20

Samples with a confirmed band (WB): A431 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Reactivity Notes

The supplier lists reactivity with human. UniProt places ubiquitin carboxyl-terminal hydrolase 28 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 122.5 kDa; observed band at about 135 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human USP28.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
