Product Overview
This rabbit polyclonal antibody recognizes ubiquitin carboxyl-terminal hydrolase 9X (gene USP9X) in human, mouse and rat samples and is validated for Western blot, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 292.3 kDa, and the supplier reports an observed band at about 290 kDa.
USP9X is a large X-linked deubiquitinating enzyme that protects many proteins from proteasomal degradation. Its substrates include SMAD4, whose deubiquitination supports TGF-β/BMP signaling, and the survival protein MCL1. USP9X is studied in development, neurodevelopment and cancer.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse, Rat) |
|---|---|
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human HeLa cells, human 293T cells, human HepG2 cells, rat brain tissue, rat liver tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): HEL cells.
Immunogen
Recombinant human USP9X fragment (Gln1175–Gln1283), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places ubiquitin carboxyl-terminal hydrolase 9X in the cytoplasm and cell projection. Tissue expression noted by UniProt (human): Widely expressed in embryonic and adult tissues. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
