Product Overview

Clone CIH-11 is a rabbit monoclonal antibody that recognizes vascular endothelial growth factor receptor 2 (gene KDR) in human samples and is validated for Western blot and immunoprecipitation. The predicted molecular weight is 151.5 kDa, and the supplier reports an observed band at about 180–250 kDa.

VEGFR2 (KDR, also called FLK1) is a receptor tyrosine kinase on endothelial cells that binds VEGF-A as well as processed VEGF-C and VEGF-D. Ligand binding triggers receptor dimerization and autophosphorylation, activating PLCγ, PI3K/AKT and MAPK signaling that drives endothelial proliferation, migration, survival and vascular permeability. VEGFR2 is the main mediator of VEGF-driven angiogenesis and a major target of anti-angiogenic cancer drugs.

Validated Applications

Western blot 1:500–1:2000
Immunoprecipitation 1:20

Samples with a confirmed band (WB): human placenta tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human VEGF Receptor 2.

Reactivity Notes

The supplier lists reactivity with human. UniProt places vascular endothelial growth factor receptor 2 in the cell junction, endoplasmic reticulum and cell membrane. Tissue expression noted by UniProt (human): Detected in cornea (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 151.5 kDa; observed bands at about 180 and 250 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human VEGF Receptor 2.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
