Product Overview

Clone 25V50 is a rabbit monoclonal antibody that recognizes vascular endothelial growth factor D (gene VEGFD) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 40.4 kDa, and the supplier reports an observed band at about 38 kDa.

VEGF-D is a growth factor of the VEGF family that binds and activates VEGFR-2 and VEGFR-3. It promotes the growth of blood and lymphatic vessels, with a particular role in lymphangiogenesis, and it is studied in tumor lymphatic spread and in vascular development.

Target Vascular endothelial growth factor D (gene VEGFD; UniProt O43915, human)

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:50

Samples with a confirmed band (WB): human Caco-2 cells, human HepG2 cells, human A549 cells, human HeLa cells, rat heart tissue, rat lung tissue, mouse heart tissue, mouse lung tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human VEGFD.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt describes vascular endothelial growth factor D as a secreted protein. Tissue expression noted by UniProt (human): Highly expressed in lung, heart, small intestine and fetal lung, and at lower levels in skeletal muscle, colon, and pancreas. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
