Vendor description (summary): Boster Bio Anti-Vinculin/VCL Antibody Picoband® catalog # A01207-1.

Biological background

Biological context: Actin filament (F-actin)-binding protein involved in cell-matrix adhesion and cell-cell adhesion. Regulates cell-surface E-cadherin expression and potentiates mechanosensing by the E-cadherin complex. May also play important roles in cell morphology and locomotion.

Expression and localization notes: cellular localization: Cell membrane. Peripheral membrane protein. Cytoplasmic side. Sarcolemma. Cytoskeleton. Adherens junction. Focal adhesion., tissue context: Metavinculin is muscle-specific..

Common research applications

  • Western blotting (WB): Compare VCL levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of VCL in tissue sections, considering fixation and antigen retrieval effects.
  • Immunofluorescence / ICC: Assess subcellular localization patterns and co-localization with compartment markers in cultured cells.
  • Flow cytometry: Quantify VCL-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: In mammalian cells, vinculin is a membrane-cytoskeletal protein in focal adhesion plaques that is involved in linkage of integrin adhesion molecules to the actin cytoskeleton. It is mapped to 10q22.2. Vinculin is a cytoskeletal protein associated with cell-cell and cell-matrix junctions, where it is thought to function as one of several interacting proteins involved in anchoring F-actin to the membrane. Defects in VCL are the cause of cardiomyopathy dilated type 1W. Dilated cardiomyopathy is a disorder characterized by ventricular dilation and impaired systolic function, resulting in congestive heart failure and arrhythmia. Multiple alternatively spliced transcript variants have been found for this gene, but the biological validity of some variants has not been determined.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cell membrane. Peripheral membrane protein. Cytoplasmic side. Sarcolemma. Cytoskeleton. Adherens junction. Focal adhesion.
  • Tissue details: Metavinculin is muscle-specific.
  • Research category: Actin Binding Proteins,Actin, etc.,Cardiovascular,Contractility,Cytoskeleton,Cytoskeleton/ECM,Microfilaments,Signal Transduction
