Product Overview
This rabbit polyclonal antibody recognizes vacuolar protein sorting-associated protein 53 homolog (gene VPS53) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 79.7 kDa, and the supplier reports an observed band at about 100 kDa.
VPS53 is a subunit of the GARP and EARP complexes, which tether vesicles in endosome-to-Golgi and endosomal recycling pathways. GARP keeps mannose-6-phosphate receptors cycling so that acid hydrolases reach lysosomes. VPS53 mutations cause a severe neurodevelopmental disorder, and it is studied in membrane trafficking.

Samples with a confirmed band (WB): human 293T cells, human HepG2 cells, human MCF-7 cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung adenocarcinoma tissue, human spleen tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): A549 cells.
Samples with confirmed staining (ICC/IF): human breast cancer tissue.
Samples with a confirmed signal (flow cytometry): U87 cells.
Immunogen
Recombinant human VPS53 fragment (Glu11–Thr699), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places vacuolar protein sorting-associated protein 53 homolog in the endosome membrane and recycling endosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
