Product Overview

This rabbit polyclonal antibody recognizes GATOR2 complex protein WDR24 in human, mouse and rat samples and is validated for Western blot and ELISA. The predicted molecular weight is 88.2 kDa, and the supplier reports an observed band at about 88 kDa.

WDR24 is a subunit of GATOR2, a complex that links amino acid availability to mTORC1 activity. When amino acids are plentiful, GATOR2 counteracts the GATOR1 complex, allowing mTORC1 to be activated at the lysosome. WDR24 is studied in nutrient sensing, cell growth and autophagy.

Target GATOR2 complex protein WDR24 (gene WDR24; UniProt Q96S15, human)
Host / Clonality / Isotype Rabbit / Polyclonal / Rabbit IgG

Validated Applications

Western blot 0.25–0.5 µg/mL (Mouse, Rat)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human 293T cells, human placenta tissue, monkey COS-7 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant human WDR24 fragment (His309–His762), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places GATOR2 complex protein WDR24 in the lysosome membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?

A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 88.2 kDa; observed band at about 88 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human WDR24 fragment (His309–His762), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
