Product Overview

Clone 25W87 is a rabbit monoclonal antibody that recognizes tRNA (gene WDR4) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 45.5 kDa, and the supplier reports an observed band at about 45 kDa.

WDR4 is a WD-repeat protein that pairs with the methyltransferase METTL1 to form the enzyme complex that places N7-methylguanosine (m7G) on RNA. WDR4 contributes no catalytic activity but holds tRNA in place, supporting m7G46 modification that stabilizes tRNA structure and also affecting certain mRNAs and microRNAs, so it is studied in RNA modification, translation control and development.

Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Flow cytometry 1:50

Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human 293T cells, rat heart tissue, rat liver tissue, mouse heart tissue, mouse liver tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human WDR4.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places tRNA in the nucleus and chromosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
