Product Overview
Clone ADHA-23 is a rabbit monoclonal antibody that recognizes WD repeat-containing protein 5 (gene WDR5) in human, mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 36.6 kDa, and the supplier reports an observed band at about 37 kDa.
WDR5 is a WD40-repeat scaffold protein found in the MLL/SET1 histone methyltransferase complexes, where together with RBBP5 and ASH2L it supports methylation of histone H3 at lysine 4, a mark of active transcription. It also belongs to the NSL acetyltransferase complex, and it is widely studied in chromatin regulation, stem cells and cancer biology.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
| Flow cytometry | 1:50 |
Samples with a confirmed band (WB): human Jurkat cells lysate, human MOLT-4 cells lysate, human MCF-7 cells lysate.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human WDR5.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places WD repeat-containing protein 5 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
