Rabbit Monoclonal

Product Overview

Clone 28W88 is a rabbit monoclonal antibody that recognizes protein Wnt-16 (gene WNT16) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 40.7 kDa, and the supplier reports an observed band at about 41 kDa.

Wnt-16 is a secreted member of the Wnt family of signaling proteins that binds Frizzled receptors. It is thought to act locally during tissue development, and genetic and animal studies have linked it to cortical bone thickness and bone strength.

Validated Applications

Western blot 1:500–1:2000

Samples with a confirmed band (WB): human SH-SY5Y cells, human U251 cells, rat kidney tissue, mouse kidney tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human wnt16.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Isoform Wnt-16b is expressed in peripheral lymphoid organs such as spleen, appendix, and lymph nodes, in kidney but not in bone marrow. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
