Product Overview
This rabbit polyclonal antibody recognizes protein Wnt-5a (gene WNT5A) in human samples and is validated for Western blot and ELISA. The predicted molecular weight is 42.3 kDa, and the supplier reports an observed band at about 43 kDa.
Wnt-5a is a secreted Wnt ligand best known for non-canonical signaling, acting through receptors such as ROR2 and Frizzled to regulate cell polarity and migration. Depending on receptor context it can either activate or oppose β-catenin signaling, and it is required for outgrowth of the body axis, limbs and genitalia, so it is studied in development, inflammation and cancer.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human SiHa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Recombinant human WNT5A fragment (Asp198–Asp303), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 100%.
Reactivity Notes
The supplier lists reactivity with human. UniProt describes protein Wnt-5a as a secreted protein. Tissue expression noted by UniProt (human): Expression is increased in differentiated thyroid carcinomas compared to normal thyroid tissue and anaplastic thyroid tumors where expression is low or undetectable. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
