Product Overview
Clone 19X78 is a rabbit monoclonal antibody that recognizes xanthine dehydrogenase/oxidase (gene XDH) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 146.4 kDa, and the supplier reports an observed band at about 146 kDa.
Xanthine dehydrogenase/oxidase (XDH) is a molybdenum-containing enzyme that catalyzes the last two steps of purine breakdown, oxidizing hypoxanthine to xanthine and xanthine to uric acid. It can exist as a dehydrogenase or be converted to an oxidase form that generates reactive oxygen species, and it is the target of urate-lowering drugs such as allopurinol, making it relevant to gout and oxidative stress research.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
Samples with a confirmed band (WB): 293T cells, rat liver tissue, mouse liver tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse small intestine tissue, rat small intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human xanthine Oxidase.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes xanthine dehydrogenase/oxidase as secreted and also located in the cytoplasm and peroxisome. Tissue expression noted by UniProt (human): Detected in milk (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
