Product Overview

Clone 25E62 is a rabbit monoclonal antibody that recognizes general transcription and DNA repair factor IIH helicase subunit XPD (gene ERCC2) in human, mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 86.9 kDa, and the supplier reports an observed band at about 73 kDa.

XPD (ERCC2) is an ATP-dependent 5′-3′ DNA helicase and subunit of the TFIIH complex, which functions in both RNA polymerase II transcription and nucleotide excision repair. In repair, XPD verifies damage and opens DNA around lesions. Mutations cause xeroderma pigmentosum group D, trichothiodystrophy and related disorders.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): human K562 cells, human U251 cells, human SiHa cells, human HepG2 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): human ovarian cancer tissue, human skin cancer tissue, mouse testis tissue, rat testis tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human XPD.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places general transcription and DNA repair factor IIH helicase subunit XPD in the nucleus and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 86.9 kDa; observed band at about 73 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human XPD.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
