Overview

This antibody is intended for detection of XRCC4 (DNA repair protein XRCC4) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-XRCC4 Antibody Picoband® catalog # PB9908. Tested in IHC, WB applications. This antibody reacts with Human. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: A synthetic peptide corresponding to a sequence at the N-terminus of human XRCC4, different from the related mouse sequence by four amino acids.
  • Molecular weight context: reported MW: 55 kDa; calculated MW: 38287 MW
  • Reactivity: Human
  • Applications: IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

DNA repair protein XRCC4; DNA repair protein XRCC4. DNA repair protein XRCC4, also known as X-ray repair cross-complementing protein 4 or XRCC4, is a protein that in humans is encoded by the XRCC4 gene. In addition to humans, the XRCC4 protein is also expressed in many other metazoans, fungi and in plants. The X-ray repair cross-complementing protein 4 is one of several coreproteins involved in the non-homologous end joining (NHEJ) pathway to repair DNA double strand breaks (DSBs). Since XRCC4 is the key protein that enables interaction of LigIV to damaged DNA and therefore ligation of the ends, mutations in the XRCC4 gene were found to cause embryonic lethality in mice and developmental inhibition and immunodeficiency in humans. Furthermore, certain mutations in the XRCC4 gene are associated with an increased risk of cancer. Functional note: Involved in DNA non-homologous end joining (NHEJ) required for double-strand break repair and V (D)J recombination. Binds to DNA and to DNA ligase IV (LIG4). The LIG4-XRCC4 complex is responsible for the NHEJ ligation step, and XRCC4 enhances the joining activity of LIG4. Binding of the LIG4-XRCC4 complex to DNA ends is dependent on the assembly of the DNA-dependent protein kinase complex DNA-PK to these DNA ends. . Reported localization: Nucleus . Expression/tissue context: Widely expressed. .

Research relevance and current trends

  • DNA/RNA: Researchers commonly examine how XRCC4 (DNA repair protein XRCC4) relates to this theme using model systems and orthogonal readouts.
  • DNA Damage & Repair: Researchers commonly examine how XRCC4 (DNA repair protein XRCC4) relates to this theme using model systems and orthogonal readouts.
  • Epigenetics and Nuclear Signaling: Researchers commonly examine how XRCC4 (DNA repair protein XRCC4) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative XRCC4 (DNA repair protein XRCC4) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of XRCC4 (DNA repair protein XRCC4) across tissue regions and cell types using matched controls.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
