Product Overview
This rabbit polyclonal antibody recognizes DNA repair protein XRCC4 in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 38.3 kDa, and the supplier reports an observed band at about 55 kDa.
XRCC4 is a core scaffold of non-homologous end joining, the main pathway that repairs DNA double-strand breaks in mammalian cells. It stabilizes DNA ligase IV and, together with XLF, forms filaments that bridge broken DNA ends so they can be rejoined, which is also essential for V(D)J recombination during lymphocyte development.

Samples with a confirmed band (WB): human HepG2 cells, human 293T cells, human A431 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung squamous cells carcinoma tissue, human breast duct carcinoma tissue, human colorectal adenocarcinoma tissue, human liver cancer tissue, human ovarian cancer tissue, human penis squamous cells carcinoma tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with confirmed staining (ICC/IF): human colorectal adenocarcinoma tissue.
Samples with a confirmed signal (flow cytometry): 293T cells.

Reactivity Notes
The supplier lists reactivity with human. UniProt places DNA repair protein XRCC4 in the nucleus and chromosome. Tissue expression noted by UniProt (human): Widely expressed. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
