Product Overview
Clone CIE-25 is a rabbit monoclonal antibody that recognizes transcriptional coactivator YAP1 in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 54.5 kDa, and the supplier reports an observed band at about 70 kDa.
YAP1 is a transcriptional coactivator and the main downstream effector of the Hippo pathway. When Hippo signaling is active, LATS1/2 kinases phosphorylate YAP1 and keep it out of the nucleus; when the pathway is off, YAP1 enters the nucleus and partners with TEAD factors to drive genes for growth and survival, so it is widely studied in organ size control, mechanotransduction, regeneration and cancer.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human 293T cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:5000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human uterus tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Immunogen
Synthetic peptide based on the sequence of human YAP1.
Reactivity Notes
The supplier lists reactivity with human. UniProt places transcriptional coactivator YAP1 in the cytoplasm, nucleus, cell junction and cell membrane. Tissue expression noted by UniProt (human): Increased expression seen in some liver and prostate cancers. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
