Product Overview
Clone DDB-25 is a rabbit monoclonal antibody that recognizes Y-box-binding protein 1 (gene YBX1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 35.9 kDa, and the supplier reports an observed band at about 48 kDa.
Y-box-binding protein 1 (YBX1) is a cold-shock domain protein that binds both DNA and RNA. It is a major component of cytoplasmic mRNPs, where it influences mRNA packaging, stability and translation, and it also takes part in splicing, DNA repair and transcription; it is often elevated in tumors and studied in cancer and stress responses.

Samples with a confirmed band (WB): human HeLa cells, human MCF-7 cells, human Jurkat cells, human T-47D cells, rat spleen tissue, rat PC-12 cells, mouse spleen tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human kidney tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Samples with confirmed staining (ICC/IF): MCF-7 cells.

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes Y-box-binding protein 1 as secreted and also located in the cytoplasm, nucleus and cytoplasmic granule. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
