Product Overview

This rabbit polyclonal antibody targets transcriptional repressor protein YY1 in human, mouse and rat samples and is validated for flow cytometry, ICC/IF and Western blot. The predicted molecular weight is 44.7 kDa, and the supplier reports an observed band at about 65 kDa.

YY1 (Yin Yang 1) is a ubiquitously expressed zinc finger transcription factor that can either activate or repress genes depending on its partners and promoter context. It binds sequences near many transcription start sites, recruits chromatin-modifying enzymes, and helps form enhancer-promoter loops, making it important in development, imprinting and cancer.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat)
Immunocytochemistry / Immunofluorescence 2 µg/mL (Human)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)

Samples with a confirmed band (WB): mouse thymus tissue, mouse spleen tissue, rat thymus tissue, rat PC-12 cells, mouse RAW264.7 cells, human PC-3 cells, human SW620 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (ICC/IF): A431 cells.

Samples with a confirmed signal (flow cytometry): THP-1 cells.

Immunogen

Synthetic peptide from the middle region of human YY1. The peptide sequence is conserved in mouse.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places transcriptional repressor protein YY1 in the nucleus matrix. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Mouse, Rat); Immunocytochemistry / Immunofluorescence 2 µg/mL (Human); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on bovine samples?
A.Reactivity with bovine samples has not been tested by the supplier. Aligning the immunogen sequence with the bovine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 44.7 kDa; observed band at about 65 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the middle region of human YY1. The peptide sequence is conserved in mouse.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
