Product Overview

This rabbit polyclonal antibody targets polycomb protein eed in zebrafish samples and is validated for Western blot.

EED is a core WD40 subunit of Polycomb repressive complex 2 (PRC2). It binds trimethylated H3K27 and stimulates the methyltransferase EZH2, helping spread this repressive mark to silence developmental genes, and it is studied in development, stem cells and cancer.

Validated Applications

Western blot 0.25–0.5 µg/mL (Zebrafish)

Samples with a confirmed band (WB): zebrafish head tissue, whole female zebrafish tissue, whole male zebrafish tissue, zebrafish embryo tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant zebrafish EED fragment (Lys79–Arg443), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with zebrafish. UniProt places polycomb protein eed in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Zebrafish). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Zebrafish. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: observed band at about 45 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant zebrafish EED fragment (Lys79–Arg443), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
