Product Overview
This rabbit polyclonal antibody targets LIM/homeobox protein Lhx3 in zebrafish samples and is validated for Western blot and IHC.
LHX3 is a LIM-homeodomain transcription factor essential for development of the pituitary gland and motor neurons. With partners such as ISL1 and LDB1, it specifies motor neuron and interneuron identity in the spinal cord, and it activates pituitary hormone genes. LHX3 mutations cause combined pituitary hormone deficiency, and zebrafish lhx3 is used to study neural development.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Zebrafish) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Zebrafish) |
Samples with a confirmed band (WB): zebrafish head tissue, whole female zebrafish tissue, whole male zebrafish tissue, zebrafish embryo tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): zebrafish brain tissue, zebrafish spinal cord tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant zebrafish LHX3 fragment (Gln11–Gln330), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with zebrafish. UniProt places LIM/homeobox protein Lhx3 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
