Product Overview

This rabbit polyclonal antibody recognizes E3 ubiquitin-protein ligase mib1 in zebrafish samples and is validated for Western blot.

Mind bomb 1 (MIB1) is an E3 ubiquitin ligase that ubiquitinates Delta and Jagged-type Notch ligands, triggering their endocytosis and enabling them to activate Notch on neighboring cells. It is therefore required for Notch signaling during development, and zebrafish mib mutants are a classic model of Notch loss. MIB1 also regulates centriolar satellite proteins and primary cilium formation.

Validated Applications

Western blot 0.25–0.5 µg/mL (Zebrafish)

Samples with a confirmed band (WB): zebrafish head tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Recombinant zebrafish MIB1 fragment (Ile78–Lys995), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with zebrafish. UniProt places E3 ubiquitin-protein ligase mib1 in the cytoplasm and cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
