Product Overview

This rabbit polyclonal antibody detects sonic hedgehog protein (gene shha) in zebrafish samples and is validated for Western blot and IHC.

In zebrafish, shha is the main sonic hedgehog gene, encoding a secreted morphogen that activates Smoothened signaling. Together with shhb it helps pattern the ear, the thalamus and prethalamus and many other embryonic tissues, and zebrafish shha is widely used to study Hedgehog signaling in development.

Validated Applications

Western blot 0.25–0.5 µg/mL (Zebrafish)
IHC (paraffin sections) 2–5 µg/mL (Zebrafish)

Samples with a confirmed band (WB): zebrafish head tissue, whole female zebrafish tissue, whole male zebrafish tissue, zebrafish embryo tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): zebrafish brain tissue, zebrafish cerebellum tissue, zebrafish muscle tissue, zebrafish colon tissue, zebrafish embryo tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Recombinant zebrafish SHHA fragment (Gln208–His409), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with zebrafish. UniProt places sonic hedgehog protein in the Golgi apparatus membrane. Tissue expression noted by UniProt (zebrafish): Expressed in the ventral midline of the neural tube and brain. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Zebrafish); IHC (paraffin sections) 2–5 µg/mL (Zebrafish). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Zebrafish. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: observed band at about 55 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant zebrafish SHHA fragment (Gln208–His409), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
