Product Overview
This rabbit polyclonal antibody targets centromere/kinetochore protein zw10 homolog in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 88.8 kDa, and the supplier reports an observed band at about 95 kDa.
ZW10 is a kinetochore protein that forms the RZZ complex with ROD and ZWILCH. During mitosis this complex recruits dynein-dynactin and MAD1–MAD2 to kinetochores, supporting the spindle assembly checkpoint that prevents premature chromosome separation; outside mitosis ZW10 also takes part in retrograde transport between the Golgi and the endoplasmic reticulum.

Samples with a confirmed band (WB): human HeLa cells, human K562 cells, human Jurkat cells, human Caco-2 cells, human HL-60 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): 293T cells.

Recombinant human ZW10 fragment (Lys35–Lys779), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places centromere/kinetochore protein zw10 homolog in the cytoplasm, endoplasmic reticulum membrane and chromosome.
