In Vitro
Antibiotic DC 81 shows cytotoxicity against B16, A375, A2058, and RPMI7951 human melanoma cell lines, with IC50 values, in that order, of 4.4 μM, 18.5 μM, 31.0 μM, and 41.5 μM[1][2]. Antibiotic DC 81 acts by covalently binding the N2 of guanine in the DNA minor groove through its electrophilic carbinolamine functionality at N10-C11[1]. At 4 μM for 24 h, Antibiotic DC 81 induces mitochondria dependent apoptosis[2].

In Vivo
In tumor-bearing mice, Antibiotic DC 81 (0-10 mg/kg, i.p.; days 4, 7, 10 and 13 after tumor cell injection) decreases the tumor burden, but at 10 mg/kg it significantly impairs cardiac muscle enzyme and liver function[2].
